Introduction
Tembusu virus (TMUV, genus Flavivirus, family Flaviviridae) was first isolated in 1955 from Culex tritaeniorhynchus mosquitoes in Kuala Lumpur, Malaysia. In April 2010, duck TMUV was first identified as the causative agent of egg-drop syndrome, characterized by a substantial decrease in egg laying and depression, growth retardation and neurological signs or death in infected egg-laying and breeder ducks, in the China.
In 2014, the first duck strain of Tembusu virus was isolated from diseased duck in the Southwest of China (TMUV CQW1 strain, GenBank Accession: KM233707). Based on that, the monoclonal antibody against E protein, the polyclonal antibody against whole virus particle, NS1, NS2A, NS3, NS4B, and NS5 were prepared. The functional TMUV C, prM, E, NS1, NS2A, NS2B, NS3, NS4B, NS5 protein were obtained. A series of TMUV pathogen detection methods were developed including real time quantitative PCR detection of TMUV E, diagnostic kit for anti-TMUV E (Colloidal Gold), et al. Currently, duck TMUV reverse genetic systems have been constructed, including: TMUV infectious cDNA clone, reporter TMUV virus, TMUV replicons, as well as TMUV single-round infectious particle. Based on those, we are working on the following area:
1. The molecular mechanism of TMUV RNA replication.
2. The immune interplay between the host protein and duck TMUV NSs.
3. Autophagy and apoptosis induced by DTMUV infection.
4. MiRNA modulated viral replication.
5. Novel vaccine development.
